L-tryptophan is generally well tolerated but has a significant past safety concern: in 1989, over 1,500 cases of eosinophilia-myalgia syndrome (EMS)—a serious neurological disorder causing skin changes, hair loss, and rash—were linked to contaminated L-tryptophan from a single Japanese manufacturer. About 95% of cases traced to that source, and the FDA restricted L-tryptophan imports in 1990.

Common mild side effects from L-tryptophan include belching, diarrhea, drowsiness, dry mouth, flatulence, headache, heartburn, lightheadedness, nausea, stomach pain, visual blurring, and vomiting. L-theanine is generally well tolerated short-term but long-term safety is not well studied; reported side effects include headache, drowsiness, increased sleep duration, and increased dream activity.

PharmaGABA is often well tolerated short-term, with reported side effects of drowsiness, gastric upset, minor throat burning, muscle weakness, and nausea, but long-term safety is unclear. None of these ingredients have adequate safety data for pregnancy or breastfeeding — the facts advise against use in both situations.
