Acetaminophen, Chlorpheniramine, Codeine, Phenylephrine falls under CNS DEPRESSANTS — the group Theanine interacts with.
Theoretically, theanine may compete with glutamate and/or increase plasma gamma-aminobutyric acid (GABA) levels, which could cause CNS depression. In one clinical study, some subjects taking oral theanine reported drowsiness.

Here's the practical takeaway with this combo cold medicine (Colrex). Two ingredients in it, the codeine and the chlorpheniramine (an older antihistamine), already tend to make people feel sleepy or foggy. Theanine is thought to nudge your brain's calming signals (like GABA) in the same direction. So on paper, stacking them could add up to a little more drowsiness, slowed thinking, or feeling 'out of it.'
That said, this is a theoretical concern based on how theanine works, not something proven in real-world use, and it's rated minor and unlikely. Most people won't notice much. Still, I'd be careful driving or doing anything that needs sharp focus until you know how you feel, and run it by your pharmacist if you take theanine regularly.

This is a combination cold and cough product, and the piece that matters here is the codeine, which has mild effects on serotonin in the body. Theanine may nudge serotonin levels too, though the research is genuinely mixed, with some studies showing a small rise and others a small drop. Honestly, the signal is weak. There are no real-world reports of theanine triggering serotonin-related problems.
So for most people, taking theanine alongside Colrex is unlikely to cause trouble. I'd be more mindful of the everyday overlap: both theanine's calming effect and the codeine plus chlorpheniramine can add to drowsiness, so go easy until you know how you feel. If you ever notice agitation, racing heart, sweating, or shakiness, check in with your pharmacist or doctor.
