Theanine and Desipramine

These two interact in 2 distinct ways. Each one is explained separately below.

Why Are There 2 Interactions?

Theanine interacts with whole categories of medication. Desipramine falls into 2 of those categories, so it can interact with Theanine in 2 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Minor).

Minor

CNS DEPRESSANTS

1 of 2

Why this applies to Desipramine

Desipramine falls under CNS DEPRESSANTS — the group Theanine interacts with.

Theoretically, theanine might have additive sedative effects when used in conjunction with CNS depressants. However, it is unclear if this concern is clinically relevant.
Theoretically, theanine may compete with glutamate and/or increase plasma gamma-aminobutyric acid (GABA) levels, which could cause CNS depression. In one clinical study, some subjects taking oral theanine reported drowsiness.

Here's the simple version: theanine, the calming amino acid from tea, may have a mild relaxing effect on the brain. Desipramine, your tricyclic antidepressant, can already make some people feel a little drowsy or foggy, especially early on. Put the two together and, in theory, that sleepy feeling could add up a bit.

Honestly though, this concern is mostly on paper. It's based on how these substances might behave, not on solid studies, and it's rated as minor and unlikely. Most folks tolerate the combo fine. Just keep an eye out for extra tiredness or grogginess, go easy until you know how you feel, and check with your pharmacist or doctor before adding theanine.

SEROTONERGIC DRUGS

2 of 2

Desipramine also falls under SEROTONERGIC DRUGS.

Clinical studies regarding the effects of L-theanine on serotonin levels are conflicting. Some studies suggest it can increase serotonin levels in the brain while others report that it may decrease them. Nevertheless, there have been no reports of l-theanine being a causative agent in serotonergic-related side effects or serotonin syndrome.
Severity Minor Likelihood Unlikely Evidence C

Interaction has been demonstrated in animal or in lab research but has been shown not to occur in humans..

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the honest picture with desipramine and theanine. Desipramine is a tricyclic antidepressant that nudges up serotonin and norepinephrine in the brain. Theanine may gently touch some of those same messengers, but the research is genuinely mixed. Some studies hint it raises serotonin, others suggest it lowers it, so we can't even say which direction it leans.

The practical bottom line: this is rated minor and unlikely, and there are no real-world reports of theanine triggering serotonin-related problems like agitation, sweating, fast heartbeat, or tremor. The evidence here is weak. Still, since you're on an antidepressant, it's worth a quick word with your pharmacist or doctor before adding theanine.

What These Ratings Mean

Every interaction above is graded three ways — here’s the key to each label.

Severity how clinically significant the interaction is
Major

Clinically significant. Avoid the combination, or use only under close professional supervision.

Moderate

Usually worth avoiding or managing — may need monitoring, a dose change, or timing apart.

Minor

Generally not serious. Be aware of it and mention it to your pharmacist.

Likelihood how strong the evidence is that the interaction actually happens in people
  • LikelyWell-controlled human studies have demonstrated the likely existence of this interaction.
  • ProbableInteraction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports.
  • PossibleInteraction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
  • UnlikelyInteraction has been demonstrated in animal or in lab research but has been shown not to occur in humans.
Evidence the quality of the studies behind the rating (graded A–D)
  • AHigh-quality meta-analysis (quantitative systematic review).
  • BCase-control study.
  • CConsensus.
  • DAnecdotal evidence.

About Theanine

What is Theanine?

Theanine
  • Anxiety and stress
  • Relaxation and calm
  • Sleep support
  • Focus and cognitive function

Theanine (usually L-theanine) is an amino acid found naturally in tea leaves that many people take to feel calmer and less stressed without strong drowsiness. Early research suggests it may help with relaxation and stress, but evidence for most uses is still limited. It appears generally well tolerated for short-term use, but check with your pharmacist or doctor before starting it.
