Sotorasib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Berberine interacts with.

Theoretically, berberine might increase serum levels of drugs metabolized by CYP3A4.
In vitro research and preliminary clinical research show that berberine moderately inhibits CYP3A4. Theoretically, use of berberine with drugs metabolized by CYP3A4 might increase drug levels and increase the risk of adverse effects.

Here's the gist with Lumakras (sotorasib) and berberine. Your body relies partly on a liver enzyme called CYP3A4 to clear sotorasib out of your system. Berberine appears to slow that enzyme down a bit. The concern is that with the cleanup crew working slower, more sotorasib could build up in your blood, which may raise your chances of side effects like nausea, diarrhea, fatigue, or changes in your liver blood tests.

This is still mostly a theoretical concern based on lab and early research, so it isn't a sure thing. Still, sotorasib is a serious cancer drug, so please don't add berberine on your own. Run it by your oncologist or pharmacist first so they can keep you safe.

About Sotorasib

What Is Sotorasib Used For?

Brand name: Lumakras

Lumakras is used to treat adults with KRAS G12C-mutated locally advanced or metastatic non-small cell lung cancer (NSCLC) who have already received at least one prior cancer treatment. This use was approved under the FDA's accelerated approval pathway based on tumor response rates, and continued approval may depend on results from confirmatory trials.

Lumakras is also used in combination with panitumumab to treat adults with KRAS G12C-mutated metastatic colorectal cancer (mCRC) who have previously received certain chemotherapy regimens. In both cases, patients must first be confirmed to carry the KRAS G12C mutation using an FDA-approved test.
