Animal research suggests that berberine may have sedative effects. Theoretically, use of berberine along with CNS depressants might produce additive therapeutic and adverse effects.

Here's the simple version: Diazepam (Valium) already calms the brain, and that's exactly what makes you feel drowsy or relaxed when you take it. Berberine, in animal studies, seems to have its own mild calming effect. So the concern is that putting the two together might add up, leaving you sleepier than expected.
In real life that could look like more drowsiness, grogginess, dizziness, or trouble concentrating. I want to be honest with you, though: this is a theoretical worry based only on animal research, not strong human data, so it may not amount to much for most people. Still, if you notice extra sleepiness, or you drive or operate machinery, mention it to your pharmacist or doctor before combining them.

CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES

Preliminary clinical research shows that berberine can inhibit CYP2C9. Theoretically, taking berberine with drugs metabolized by CYP2C9 might increase drug levels and increase the risk of adverse effects.

Here's the gist if you're thinking about pairing berberine with diazepam (Valium). Your liver uses several enzymes to clear diazepam, and one of them is called CYP2C9. Early research hints that berberine can slow that enzyme down. If diazepam gets cleared a little slower, more of it can linger in your system, which could nudge up its calming effects, things like extra drowsiness, dizziness, grogginess, or feeling unsteady on your feet.
I want to be honest about the evidence here. This is mostly theoretical and based on limited, lower-quality research, so it may not amount to much for many people. Still, since both can make you sleepy, I'd run it past your pharmacist or doctor before combining them rather than starting berberine on your own.

Here's the gist for Valium (diazepam): your liver uses an enzyme called CYP3A4 to help break it down and clear it out. Berberine appears to slow that same enzyme down a bit. When that happens, diazepam can hang around longer than usual, so its calming effects may stack up more than you'd expect.

What Is Diazepam Used For?

Diazepam treats several different problems, depending on the product. Oral diazepam (tablets such as Valium, and oral solution) is used for anxiety disorders, short-term relief of anxiety symptoms, and symptom relief in acute alcohol withdrawal. It is also used alongside other treatments for muscle spasm and, adjunctively, for convulsive disorders (seizure disorders).
Diazepam injection is used for the same anxiety and alcohol withdrawal uses. It is also used before endoscopic procedures, before surgery, and before cardioversion (when given intravenously). It helps with muscle spasm and tetanus, and with status epilepticus (a prolonged, dangerous seizure) as an add-on. Diazepam rectal gel and Valtoco nasal spray are used only for seizure clusters (acute repetitive seizures) in people with epilepsy ages 2 and older.
