Larotrectinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Clove interacts with.

Theoretically, concomitant use of clove may increase levels of drugs metabolized by CYP3A4.
In vitro research shows that eugenol, the principal constituent of clove, can inhibit CYP3A4 in a dose-dependent manner. This effect has not been reported in humans.

Your body relies on a liver enzyme called CYP3A4 to break down larotrectinib (Vitrakvi) and clear it out. Clove's main compound, eugenol, has been shown in lab dishes to slow that same enzyme down. If that happened in people, larotrectinib could build up in your blood, which might mean stronger side effects like dizziness, fatigue, nausea, or changes in your liver tests.

Here's the reassuring part: this concern comes only from test-tube and animal research, and it has not been seen in actual people. Sprinkling clove on food almost certainly won't matter. Concentrated clove oil or clove supplements are the bigger question mark. Since larotrectinib treats serious cancer, please run any clove supplements past your oncologist or pharmacist first.

What Is Larotrectinib Used For?

Brand name: Vitrakvi

Vitrakvi is used to treat adult and pediatric patients with solid tumors that have a neurotrophic receptor tyrosine kinase (NTRK) gene fusion — a specific genetic alteration — without a known acquired resistance mutation. It is used when the tumor has spread (is metastatic) or when surgery would likely cause severe harm, and when there are no satisfactory alternative treatments or the cancer has continued to grow after prior treatment.

Patients are selected for Vitrakvi based on an FDA-approved test that confirms the NTRK gene fusion in tumor tissue. This approval was granted under the FDA's accelerated approval pathway, based on overall response rate and duration of response; continued approval may depend on results from confirmatory trials.
