Simeprevir Sodium falls under CYTOCHROME P450 3A4 (CYP3A4) INHIBITORS — the group Ergot interacts with.

Ergotamine, a constituent of ergot, is a substrate of cytochrome P450 3A4 (CYP3A4). Theoretically, drugs that inhibit CYP3A4 might increase the risk of ergot toxicity. Some of these drugs include amiodarone (Cordarone), clarithromycin (Biaxin), diltiazem (Cardizem), erythromycin (E-mycin, Erythrocin), indinavir (Crixivan), ritonavir (Norvir), saquinavir (Fortovase, Invirase), and many others.

Here's the gist for Olysio (simeprevir): it gently slows down a liver enzyme called CYP3A4, and that same enzyme is what your body uses to break down ergot's active chemicals (like ergotamine). When that cleanup crew is slowed, ergot can build up and stick around longer than it should.

That matters because ergot already powerfully tightens blood vessels. Too much can cause cold, numb, or painful hands and feet, leg cramping, headaches, or in serious cases reduced blood flow to the limbs. Honestly, raw ergot isn't safe as a home remedy at all. This particular concern rests on theoretical and anecdotal evidence, so it's a flag, not a panic button. Please run any ergot product past your pharmacist or doctor first.

Ergot is a toxic fungus that grows on rye and contains powerful chemicals (ergot alkaloids) that strongly narrow blood vessels and squeeze the uterus. Some purified ergot chemicals are used as prescription drugs under strict medical supervision, but the crude herb is not safe to take.

What Is Simeprevir Used For?

Brand name: Olysio

  • Simeprevir is used along with ribavirin (Copegus, Rebetol) and peginterferon alfa (Pegasys) to treat chronic hepatitis C (an ongoing viral infection that damages the liver). Simeprevir is in a class of medications called protease inhibitors. It works by decreasing the amount of hepatitis C virus (HCV) in the body. Simeprevir may not prevent the spread of hepatitis C to other people.
