Promethazine falls under ANTICHOLINERGIC DRUGS — the group Huperzine A interacts with.
Huperzine A has acetylcholinesterase (AChE) inhibiting effects. In animal models, huperzine A reversed cognitive deficits induced by scopolamine, an anticholinergic drug.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the simple version. Promethazine (Phenergan) works partly by blocking acetylcholine, the brain chemical that helps with things like memory and movement. Huperzine A does the opposite, it raises acetylcholine levels by slowing the enzyme that breaks it down. So in theory, the two can work against each other, and huperzine A could blunt some of what promethazine is doing.
The honest truth is this concern comes from lab and animal studies, not real people, so it's more of a theoretical worry than a proven problem. If you do notice promethazine isn't calming your nausea or allergy symptoms as well, that's worth mentioning. Before pairing these, just run it by your pharmacist or doctor so you're not guessing.

Huperzine A is a purified compound from a Chinese clubmoss that acts like a mild cholinesterase inhibitor, similar in mechanism to some prescription Alzheimer's drugs. Some small studies suggest it may modestly help memory in people with dementia, but the evidence is limited and the quality is uncertain. What Is Promethazine Used For?
Brand name: Phenergan
Promethazine is used for a wide range of conditions. Tablets, oral solution, and rectal suppositories (brands including Phenergan, Promethegan, and Phenadoz) are approved for seasonal and perennial allergic rhinitis, vasomotor rhinitis, allergic conjunctivitis, hives (urticaria), angioedema, dermographism, and allergic reactions to blood or plasma. They are also used for motion sickness (both prevention and active treatment), nausea and vomiting related to anesthesia and surgery, sedation, and as a helper medication alongside pain relievers after surgery.
Promethazine hydrochloride injection (brands including Phenergan and Promethazine HCl injection) is used for many of the same purposes but is reserved for situations where oral treatment is not possible — including allergic emergencies as an adjunct to epinephrine, sedation during labor, and as an aid during certain surgical procedures such as bronchoscopy and eye surgery.
