CNS DEPRESSANTS

Why this applies to Meprobamate

Theoretically, concomitant use of L-tryptophan with CNS depressants might cause additive sedative effects.
Clinical research shows that L-tryptophan can cause fatigue and drowsiness.
Severity Major Likelihood Probable Evidence B

Interaction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports..

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the simple version: meprobamate already works by calming brain activity to ease anxiety, so it naturally makes many people feel relaxed, drowsy, or a bit foggy. L-tryptophan can add its own sleepy effect because your body turns it into serotonin and melatonin, the chemicals that nudge you toward sleep. Stacked together, those two can pile on, leaving you more sedated than you expect.

In real life that might look like extra grogginess, heavy drowsiness, slowed reactions, or feeling unsteady on your feet. This is rated a major interaction, though the additive sedation part is largely theory plus the known fact that tryptophan causes fatigue. Don't start or stop either one on your own. Please check with your pharmacist or doctor first, especially before driving.

About Meprobamate

What Is Meprobamate Used For?

Brand names: Equanil, Miltown, Meprospan, Neuramate

Meprobamate tablets are used to manage anxiety disorders or to provide short-term relief of anxiety symptoms. It is intended for anxiety that rises to the level of a disorder — everyday stress and tension that most people experience generally do not require this type of medication.

Because its effectiveness beyond four months has not been well studied, meprobamate is typically used for a limited period of time, with a prescriber checking in regularly to decide whether continued use is still appropriate.
