L-tryptophan is a precursor to serotonin. Theoretically, combining serotonergic drugs with L-tryptophan might increase the risk of serotonergic side effects, including serotonin syndrome and cerebral vasoconstrictive disorders.

Here's the thing about tapentadol (Nucynta): besides its opioid pain action, it also boosts serotonin in your nervous system. L-tryptophan is a building block your body turns into serotonin. So when you pair the two, you're essentially raising serotonin from two directions at once, and that's where the concern comes in.

In theory, this combo could nudge up your risk of serotonin syndrome. Watch for things like agitation, a racing heart, sweating, shakiness or muscle twitching, fever, or confusion. To be fair, this is based on how these substances work rather than strong real-world studies, so it's a possible risk, not a sure thing. Still, please check with your pharmacist or doctor before combining them, and don't change your tapentadol on your own.

About Tapentadol

What Is Tapentadol Used For?

Brand names: Nucynta, Nucynta ER

Tapentadol tablets (including Nucynta and Tapentadol Hydrochloride tablets) are used for severe acute pain in adults when other treatments are not adequate. Nucynta tablets are also approved for acute pain in children aged 6 and older who weigh at least 40 kg.

Nucynta ER and tapentadol extended-release tablets are used for severe, ongoing pain that requires around-the-clock opioid treatment and cannot be managed with other options, including severe neuropathic pain (nerve pain) caused by diabetic peripheral neuropathy in adults. Extended-release formulations are not intended for as-needed use.
