Resveratrol and Estradiol, Medroxyprogesterone

These two interact in 2 distinct ways. Each one is explained separately below.

Why Are There 2 Interactions?

Resveratrol interacts with whole categories of medication. Estradiol, Medroxyprogesterone falls into 2 of those categories, so it can interact with Resveratrol in 2 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).

Moderate

CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES

1 of 2

Why this applies to Estradiol, Medroxyprogesterone

Estradiol, Medroxyprogesterone falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Resveratrol interacts with.

Theoretically, resveratrol might increase levels of drugs metabolized by CYP1A2.
In vitro research shows that resveratrol can inhibit CYP1A2 enzymes. However, this interaction has not been reported in humans.
Severity Moderate Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

If you take this combined hormone product (estradiol plus medroxyprogesterone, sold as Divitren), here's the gist. Resveratrol has been shown in test-tube studies to slow down a liver enzyme called CYP1A2, which is one of the pathways that helps break down estradiol. In theory, blocking that enzyme could let a little more estrogen linger in your system, which might bump up estrogen-type effects like breast tenderness, bloating, nausea, headaches, or spotting.

The reassuring part: this is only based on lab work, not real people, so it's quite theoretical. Everyday food amounts from grapes or berries are very unlikely to matter; concentrated supplements are the bigger question. Before adding a resveratrol supplement, just check with your pharmacist or doctor so they can keep an eye on things.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES

2 of 2

Estradiol, Medroxyprogesterone also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.

Theoretically, resveratrol might increase levels of drugs metabolized by CYP3A4.
In vitro research shows that resveratrol can inhibit the CYP3A4 enzyme. However, clinical research shows that taking resveratrol 3000 mg daily for 8 weeks does not necessitate dose adjustments to medications metabolized by CYP3A4.

Here's the gist for your hormone combo (estradiol plus medroxyprogesterone). Both hormones are partly cleared by a liver enzyme called CYP3A4. In test-tube studies, resveratrol can slow that enzyme down a bit, which in theory could let these hormones build up slightly and bump up typical estrogen-related effects like breast tenderness, bloating, nausea, headaches, or mood changes.

The good news: this concern comes from lab work, not real patients. In fact, one human study using a hefty 3000 mg of resveratrol daily for 8 weeks did not require any dose changes for CYP3A4 medicines. So a real-world problem here is unlikely. Still, since everyone is different, it's worth a quick check with your pharmacist or doctor before adding resveratrol.

What These Ratings Mean

Every interaction above is graded three ways — here’s the key to each label.

Severity how clinically significant the interaction is
Major

Clinically significant. Avoid the combination, or use only under close professional supervision.

Moderate

Usually worth avoiding or managing — may need monitoring, a dose change, or timing apart.

Minor

Generally not serious. Be aware of it and mention it to your pharmacist.

Likelihood how strong the evidence is that the interaction actually happens in people
  • LikelyWell-controlled human studies have demonstrated the likely existence of this interaction.
  • ProbableInteraction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports.
  • PossibleInteraction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
  • UnlikelyInteraction has been demonstrated in animal or in lab research but has been shown not to occur in humans.

About Resveratrol

What is Resveratrol?

Resveratrol
  • Heart and blood vessel health
  • Antioxidant support
  • Anti-aging
  • Blood sugar and metabolic support
  • General anti-inflammatory support

Resveratrol is a plant compound found in red grapes, berries, and peanuts that is popular for heart health, anti-aging, and antioxidant support. While lab and animal studies are promising, strong human evidence for most uses is still limited. It appears generally well tolerated in supplement doses, but talk with your pharmacist or doctor before starting it.
