Why Are There 2 Interactions?
Valerian interacts with whole categories of medication. Clomipramine falls into 2 of those categories, so it can interact with Valerian in 2 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Minor).
CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES

Clomipramine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Valerian interacts with.
Although some in vitro evidence suggests that valerian affects CYP2D6, clinical pharmacokinetic (PK) studies show that valerian is unlikely to affect the CYP2D6 enzyme. In one PK study, taking valerian 1000 mg (providing about 11 mg valerenic acid) nightly for 14 days did not affect the metabolism of dextromethorphan, a CYP2D6 substrate. In another PK study, taking valerian 125 mg three times daily for 28 days did not affect metabolism of debrisoquine, an accepted CYP2D6 probe-substrate.
Interaction has been demonstrated in animal or in lab research but has been shown not to occur in humans..
Here's the good news on this one. Your body uses a liver enzyme called CYP2D6 to help clear clomipramine (Anafranil), and on paper valerian was once thought to slow that enzyme down. But when researchers actually tested valerian in people, it didn't meaningfully change how this enzyme works. So valerian is unlikely to make clomipramine build up in your system.
That said, both valerian and clomipramine can have a calming, sedating effect. Stacking them could leave you feeling extra drowsy, groggy, or foggy the next day. It's a mild concern, not a dangerous one, but it's still worth a quick chat with your pharmacist or doctor before you pair them, especially when you're driving or just starting out.

Although some in vitro evidence suggests that valerian extract might inhibit or induce CYP3A4, clinical pharmacokinetic (PK) studies show that valerian does not have a clinically significant effect on the CYP3A4 enzyme. In one PK study, taking valerian 125 mg three times daily for 28 days did not affect metabolism of midazolam, an accepted CYP3A4 probe-substrate. In another PK study, taking valerian 1000 mg (providing about 11 mg valerenic acid) nightly for 14 days modestly increases levels of alprazolam, a CYP3A4 substrate, suggesting mild inhibition of CYP3A4. However, this mild inhibition is unlikely to be clinically relevant.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the short version: your body partly uses an enzyme called CYP3A4 to clear clomipramine, and valerian was once thought to slow that enzyme down. But when researchers actually tested this in people, valerian had little to no real effect on CYP3A4 drugs. So I wouldn't expect valerian to meaningfully raise your clomipramine levels through that pathway.
That said, I'd still keep an eye on the bigger picture. Both clomipramine and valerian can make you drowsy and relaxed, so combining them may leave you more sleepy, foggy, or lightheaded than usual. That overlap, not a true blood-level change, is the part worth watching. Before adding valerian, check with your pharmacist or doctor so they can review your full medication list.
