Sildenafil falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Vitamin E interacts with.

Theoretically, vitamin E might induce metabolism of CYP3A4, possibly reducing the levels CYP3A4 substrates.
Vitamin E appears to bind with the nuclear receptor, pregnane X receptor (PXR), which results in increased expression of CYP3A4. Although the clinical significance of this is not known, use caution when considering concomitant use of vitamin E and other drugs affected by these enzymes.

Here's the gist for sildenafil (Viagra or Revatio) and vitamin E. Your body clears sildenafil using a liver enzyme called CYP3A4. In lab studies, vitamin E seems to nudge that enzyme to work a little faster. If that happens in real life, your body might break down sildenafil a bit quicker, which could make it slightly less effective. For erectile dysfunction that might mean a weaker response, and for pulmonary hypertension it could mean less benefit.

I want to be straight with you though: this is theoretical, based on how things behave in the lab, not on real patient problems. Everyday food amounts of vitamin E are very unlikely to matter. If you take high-dose supplements, just mention it to your pharmacist or doctor.

What Is Sildenafil Used For?

Brand name: Revatio

Sildenafil treats two distinct conditions depending on the product. Viagra (tablet) and Vybrique (dissolving oral film) are approved for erectile dysfunction (ED) — difficulty getting or keeping an erection. Generic sildenafil tablets and oral film are also approved for ED.

Revatio and several generic sildenafil products (tablets, oral suspension, and injection) are used for pulmonary arterial hypertension (PAH) — high blood pressure in the arteries of the lungs — in adults to improve exercise ability and slow the disease from getting worse. Revatio oral suspension and injection are also approved for PAH in children ages 1 to 17.
