L-Glutamine is generally well tolerated orally, though it can cause belching, bloating, constipation, diarrhea, flatulence, nausea, vomiting, gastrointestinal pain, headache, and musculoskeletal pain. At high doses (10–30 grams daily), digestive side effects are more common.

People with kidney or liver disease should only use it under medical supervision. Glutamine is rated likely safe in pregnancy and lactation.
