Why this applies to Gabapentin enacarbil
Gabapentin enacarbil falls under ANTICONVULSANTS — the group Glutamine interacts with.
Glutamine is metabolized to the excitatory neurotransmitter glutamate. Glutamate might have antagonistic effects with anticonvulsant drugs. However, this interaction has not yet been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the idea behind this one. Your body can turn glutamine into glutamate, which is a chemical that revs up brain activity. Horizant (gabapentin enacarbil) works in the opposite direction, helping calm overactive nerve signals to ease restless legs or shingles-related nerve pain. So in theory, loading up on glutamine could nudge against what your medication is trying to do, possibly making it work a little less well.
I want to be honest with you though: this is a theoretical concern based on how these molecules behave, not something that has actually been reported in people taking them together. There's no proven failure of seizure or pain control from glutamine. If you're using Horizant for an important reason, just give your pharmacist or doctor a quick heads-up before adding a glutamine supplement so we can keep an eye on things.

Gabapentin products are used for postherpetic neuralgia, which is nerve pain that lingers after shingles. Several gabapentin products (capsules, oral solution and tablets, including Neurontin, Relgaabi and Gabarone) are also used as add-on therapy for partial onset seizures in adults and children 3 years and older with epilepsy.
Horizant (an extended-release tablet) is used for moderate-to-severe primary restless legs syndrome in adults, plus postherpetic neuralgia. Gralise and once-daily gabapentin tablets are used only for postherpetic neuralgia.
