Moderate evidence
Glycine is rated possibly effective for schizophrenia. For other uses — including 3-phosphoglycerate dehydrogenase (3-PGDH) deficiency, ADHD, benign prostatic hyperplasia (BPH), and cystic fibrosis — the evidence we hold is insufficient to rate it.
L-theanine is rated possibly effective for cognitive function. For age-related cognitive decline, Alzheimer disease, anxiety disorders, OCD, and cancer, the evidence we hold is insufficient to rate it.

How safe is it?
Well-documented data
Both ingredients are generally well tolerated in typical short-term use, but long-term safety data are limited for glycine and not well studied for L-theanine. Glycine has rarely caused mild sedation, and some people report irritability, insomnia, fatigue, memory problems, headache, and sensory changes — though it's unclear if glycine is responsible.
Soft stools, nausea, vomiting, GI discomfort, and dry mouth have also been reported rarely. L-theanine may cause headaches, drowsiness, longer sleep, or vivid dreams.
One child on a much higher dose (400 mg daily) developed facial tics shortly after starting, but the tics stopped when theanine was discontinued — and because that child had a history of tics, a clear link wasn't established. For pregnancy, the safety data advises against use of L-theanine.
For glycine and lactation, there isn't enough data to know either way — talk with your doctor or pharmacist for personalized guidance on either point.

L-Theanine Suntheanine
Antihypertensive Drugs
Theanine might lower blood pressure, potentiating the effects of antihypertensive drugs.
Animal research shows that theanine can lower blood pressure in spontaneously hypertensive animals. Theoretically, concomitant use of theanine and antihypertensive drugs might potentiate the antihypertensive activity.
Cns Depressants
Theoretically, theanine might have additive sedative effects when used in conjunction with CNS depressants. However, it is unclear if this concern is clinically relevant.
Theoretically, theanine may compete with glutamate and/or increase plasma gamma-aminobutyric acid (GABA) levels, which could cause CNS depression. In one clinical study, some subjects taking oral theanine reported drowsiness.
Serotonergic Drugs
Clinical studies regarding the effects of L-theanine on serotonin levels are conflicting. Some studies suggest it can increase serotonin levels in the brain while others report that it may decrease them. Nevertheless, there have been no reports of l-theanine being a causative agent in serotonergic-related side effects or serotonin syndrome.
Glycine
Clozapine (Clozaril)
Theoretically, glycine might decrease the effectiveness of clozapine.
One small clinical study in patients with schizophrenia shows that adding glycine to clozapine therapy worsens symptoms of schizophrenia when compared with clozapine alone. The mechanism of this interaction is unclear.
