Our pharmacy team’s full take, with four database checks built into the cards below — a summary of what is known, not a grade of the product itself.
What’s inside
Full disclosure
This product contains a single active ingredient: L-Theanine, an amino acid naturally found in tea that's thought to support relaxation and mental clarity. The capsule also contains inactive ingredients — rice flour, microcrystalline cellulose, hypromellose, carnauba wax, silicon dioxide, and a rice extract blend — which are fillers and binders that give the supplement its form.
Does it work?
Not established
L-Theanine is possibly effective for cognitive function based on available evidence. For age-related cognitive decline, Alzheimer disease, generalized anxiety disorder, obsessive-compulsive disorder, and cancer, the evidence we have isn't reliable enough to rate it one way or the other.
If you're considering it for any of those latter uses, talk with your doctor or pharmacist about what the current research actually supports for your situation.
How safe is it?
Well-documented data
L-Theanine is generally well tolerated in healthy adults when used short-term, but long-term safety hasn't been well studied. The most commonly reported side effects are headaches and drowsiness, and some people have noticed increased sleep duration or more vivid dreams.
The safety data available doesn't support use during pregnancy or breastfeeding, so if you're pregnant or nursing, it's best to avoid this supplement and talk with your doctor or pharmacist about what might work for you instead.
Meds to double-check
Moderate interaction found
Before you take this, double-check all blood pressure medications (the interaction is moderate) and any sedating drugs — sleep aids, anxiety medications, opioids, or anything that makes you drowsy. Additionally, if you take serotonergic antidepressants, mention this product to your pharmacist so they can flag it in your records, though the interaction appears minor and unproven in practice.
The bottom line
Scorecard at a glanceFully disclosed formula with no established evidence rating for its marketed use. Moderate medication interactions have been identified, and safety information is well characterized.
L-Theanine may be worth considering if you're looking for support with mental focus and you don't take blood pressure or sedating medications. If you do take either of those drug types — or any other prescription medication — check your exact medications with the tool on this page before starting.

L-Theanine 100 mg by The Vitamin Shoppe Drug Interactions
HelloPharmacist Interaction Report
L-Theanine 100 mg by The Vitamin Shoppe interacts with blood pressure medications and sedating drugs, with the most serious being a moderate interaction with antihypertensive drugs.
L-Theanine can lower blood pressure on its own, and when combined with blood pressure medications, this effect may be amplified — potentially lowering your pressure more than intended. If you take any blood pressure medication, you'll want to check it against this product before starting.
Read the full breakdown — every affected drug type, severity by severity
L-Theanine may also have additive sedative effects when used with CNS depressants (sleep aids, anti-anxiety medications, opioids, and similar drugs that slow the central nervous system), though the clinical significance of this interaction isn't fully clear. There's also a minor theoretical interaction with serotonergic drugs (antidepressants that work on serotonin, like SSRIs), but clinical studies conflict on whether L-Theanine actually raises or lowers serotonin, and no cases of serotonin syndrome have been reported with it.
Altogether, these interactions span 565 individual medications. Use the interaction checker on this page to run your exact medications against this product before you start.

For each ingredient in L-Theanine 100 mg with known interactions, here are the types of medications it can affect. Open any type for the detail — or search your exact drug in the checker above.
L-Theanine
Antihypertensive Drugs
Theanine might lower blood pressure, potentiating the effects of antihypertensive drugs.
Animal research shows that theanine can lower blood pressure in spontaneously hypertensive animals. Theoretically, concomitant use of theanine and antihypertensive drugs might potentiate the antihypertensive activity.
Cns Depressants
Theoretically, theanine might have additive sedative effects when used in conjunction with CNS depressants. However, it is unclear if this concern is clinically relevant.
Theoretically, theanine may compete with glutamate and/or increase plasma gamma-aminobutyric acid (GABA) levels, which could cause CNS depression. In one clinical study, some subjects taking oral theanine reported drowsiness.
Serotonergic Drugs
Clinical studies regarding the effects of L-theanine on serotonin levels are conflicting. Some studies suggest it can increase serotonin levels in the brain while others report that it may decrease them. Nevertheless, there have been no reports of l-theanine being a causative agent in serotonergic-related side effects or serotonin syndrome.
