Mega-Polyzyme contains 5 active ingredients: amylase, papain, bromelain, protease, and lipase. These are all digestive enzymes — they break down different foods (carbs, proteins, and fats) to help your body absorb nutrients.
The product also contains inactive ingredients: cellulose, rice flour, and two flow agents (NU-Mag and Nu-Flow).

The evidence on whether these enzymes work for their intended purpose isn't established in the data we hold. Papain has been studied for cancer, dental cavities, shingles, muscle soreness, and parasitic infections, but the evidence for all of these is insufficient to rate.
Bromelain shows the same picture — hay fever, burns, cancer, nerve damage, circulation, and diabetes all lack reliable evidence. Lipase data shows insufficient evidence for inflammatory bowel disease and indigestion.
We simply don't have strong proof these ingredients deliver measurable benefits.
How safe is it?
Well-documented data
Papain and bromelain are generally well tolerated short-term at normal doses, though both can cause digestive upset (gas, diarrhea, cramping) and allergic reactions in sensitive people. High-dose papain carries rare but serious risks — esophageal perforation and severe inflammation of the stomach lining.
Both papain and bromelain should be avoided in pregnancy; bromelain should also be avoided while breastfeeding due to insufficient safety data. Papain has not been adequately studied in breastfeeding — talk with your doctor or pharmacist before using it.
Protease and lipase are generally well tolerated, though protease may trigger allergic reactions in sensitive individuals. Lipase has no reported adverse effects in adults, but safety data on the supplement form in pregnancy and breastfeeding is scant — use only under medical guidance.

Bromelain
Anticoagulant/Antiplatelet Drugs
Bromelain may have antiplatelet effects and may increase the risk of bleeding if used with anticoagulant or antiplatelet drugs.
There is one case report of a patient experiencing minor bruising while taking bromelain with naproxen. Bromelain is thought to have antiplatelet activity. Whether this interaction is of concern with topical bromelain is unclear. Interference with coagulation of burn wounds has been reported in a patient receiving bromelain-based enzymatic debridement. However, observational research has found that topical bromelain debridement is not associated with increases or decreases in laboratory markers of coagulation when compared with surgical debridement.
Tetracycline Antibiotics
Theoretically, bromelain might increase levels of tetracycline antibiotics.
Laboratory research suggests that bromelain might increase the absorption of tetracycline antibiotics. However, a study in healthy adults reported no difference in tetracycline plasma levels when a 500 mg dose was taken with or without bromelain 80 mg.
Papain
Warfarin (Coumadin)
Theoretically, papain might increase the effects and side effects of warfarin.
In one case report, a patient previously stable on warfarin was found to have an international normalization ratio (INR) of 7.4, which was attributed to ingestion of a supplement containing papain from papaya extract.
