Entrectinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Acacia Rigidula interacts with.

Theoretically, Acacia rigidula products that are adulterated with beta-methylphenylethylamine (BMPEA) might increase the levels of CYP3A4 substrates.
In vitro research shows that BMPEA, an adulterant found in many Acacia rigidula products, inhibits CYP3A4 enzyme by 37%.

Here's the gist with entrectinib (Rozlytrek): your body clears this cancer drug mainly through a liver enzyme called CYP3A4. Some Acacia rigidula products have been found spiked with a stimulant called BMPEA, and in lab tests that ingredient slowed CYP3A4 down by about a third. If that same thing happened in a person, entrectinib could build up higher than intended, which may mean more side effects like dizziness, a faster or irregular heartbeat, fatigue, swelling, or changes on heart tracing tests.

I want to be straight with you, though: this is only a theoretical concern based on test-tube data, not real patient studies. Still, entrectinib is a serious drug, and these supplements have real quality problems. Please don't add Acacia rigidula without first checking with your oncologist or pharmacist.

About Entrectinib

What Is Entrectinib Used For?

Brand name: Rozlytrek

Rozlytrek (entrectinib) is used to treat adult patients with ROS1-positive metastatic non-small cell lung cancer (NSCLC), as confirmed by an FDA-approved test. It is also approved for adult and pediatric patients older than 1 month of age who have solid tumors — any cancer type — that carry an NTRK gene fusion detected by an FDA-approved test, provided the tumor has no known acquired resistance mutation, is metastatic or cannot be surgically removed without severe harm, and has progressed after prior treatment or has no satisfactory alternative therapy.

The NTRK gene fusion approval in solid tumors was granted under accelerated approval, meaning it is based on tumor response rates. Continued approval may depend on results from confirmatory clinical trials.
