Acacia Rigidula and Lemborexant

Here is the interaction we found between these two, with the evidence behind it.

Moderate

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES

Why this applies to Lemborexant

Lemborexant falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Acacia Rigidula interacts with.

Theoretically, Acacia rigidula products that are adulterated with beta-methylphenylethylamine (BMPEA) might increase the levels of CYP3A4 substrates.
In vitro research shows that BMPEA, an adulterant found in many Acacia rigidula products, inhibits CYP3A4 enzyme by 37%.

Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the gist for Dayvigo (lemborexant): your body clears this sleep medicine mainly through a liver enzyme called CYP3A4. Some Acacia rigidula products have been found spiked with a stimulant called BMPEA, and in lab testing BMPEA slowed that same enzyme down by about a third. If that happens in real life, lemborexant could build up more than expected.

What that might feel like is a stronger, longer hangover effect the next day: extra grogginess, sleepiness, trouble concentrating, or feeling unsteady on your feet. I'll be honest though, this is theoretical and comes only from test-tube work, so we don't know how much it matters in people. There's also a real catch: a stimulant in a sleep aid works against each other and raises safety concerns. Please run any Acacia product past your pharmacist or doctor first.

From our pharmacist’s overview on the Lemborexant page

“Lemborexant (Dayvigo) works by quieting the brain's wake signals, so the biggest risks on this list come from anything else that sedates you. Alcohol sits at the top; a drink layered over a dose can deepen drowsiness into real impairment, and a few entries carry the added danger of slowed breathing.”

About Acacia Rigidula

What is Acacia Rigidula?

Acacia Rigidula
  • Weight loss
  • Energy and stimulation
  • Boosting metabolism
  • Appetite control
  • Pre-workout/athletic performance

Acacia rigidula is a shrub from Texas and Mexico that is marketed in weight-loss and energy supplements. There is very little reliable human evidence that it works, and some products have been found to contain an unapproved stimulant drug (BMPEA), which raises real safety concerns. Most people should avoid it and talk to a pharmacist or doctor before trying any product containing it.
