Acetaminophen, Aspirin, Caffeine falls under METHYLXANTHINES — the group Adenosine interacts with.
The methylxanthines, aminophylline, caffeine, and theophylline, can block the effects of adenosine by acting as competitive antagonists at adenosine cell surface receptors. It is recommended that methylxanthines be avoided for 24 hours prior to cardiac stress tests.

Here's the key thing about products like Excedrin: the ingredient that matters for this interaction is the caffeine. Caffeine belongs to a group called methylxanthines, and they essentially sit on the same receptors that adenosine works through and block it. So if you took an adenosine supplement, the caffeine in your headache pill could blunt some of what the adenosine is trying to do.
In everyday life this is mostly a theoretical concern based on how these compounds behave, and it's rated as minor. The acetaminophen and aspirin pieces aren't really involved. Where it genuinely matters is medical testing: caffeine is told to be avoided for about 24 hours before a cardiac stress test that uses adenosine, since it can interfere with the results.
