Lofepramine is a tricyclic antidepressant, and like many in its class it can already make some people feel drowsy or a bit foggy, especially when you first start it. The concern here is simple: berberine has shown mild sedative effects in animal studies, so in theory pairing it with lofepramine could add to that calming or sleepy feeling.

I want to be honest that this is a theoretical concern only, based on lab and animal data, not real-world human reports. For most people it is unlikely to cause trouble, but if you notice extra tiredness, dizziness, or trouble concentrating after adding berberine, that is worth mentioning. Please don't change either one on your own. A quick chat with your pharmacist or doctor is the safest way to sort it out.

Lofepramine also falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES.

Theoretically, berberine might increase serum levels of drugs metabolized by CYP2D6.
In vitro research and preliminary clinical evidence show that berberine can inhibit CYP2D6. Theoretically, use of berberine with drugs metabolized by CYP2D6 might increase drug levels and increase the risk of adverse effects.

Lofepramine is an antidepressant that your body clears largely through a liver enzyme called CYP2D6. Berberine appears to slow that same enzyme down a bit. So if you take them together, your lofepramine could clear out a little more slowly and build up higher than expected.

In real terms, that might mean more of the usual side effects you'd see with this kind of medicine: dry mouth, constipation, blurred vision, drowsiness, dizziness when you stand up, or a faster heartbeat. To be fair, this concern is mostly theoretical and based on lab work and early studies, so it may not cause trouble for everyone. Still, please don't start berberine without first checking with your pharmacist or doctor, who can keep an eye on how you're feeling.
