Here is the interaction we found between these two, with the evidence behind it.
ORAL DRUGS
Why this applies to Protriptyline
Protriptyline falls under ORAL DRUGS — the group Carob interacts with.
Laboratory research shows that carob, when consumed as part of the diet or used as a thickening agent in infant formula, reduces the absorption of certain minerals. Take carob 30-60 minutes after oral medications.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the simple version: carob is rich in fiber and tannins, and in lab studies those compounds can grab onto things in your gut and keep them from being fully absorbed. The concern with protriptyline (Vivactil) is that taking the two at the same time might let a little less of your medicine soak in. If that happened, your depression treatment could feel a touch less effective over time.
I want to be honest about how soft this evidence is. It comes only from lab and animal research, the interaction is rated minor, and it's mostly a worry with concentrated carob or carob-thickened formula, not a sprinkle in a recipe. An easy fix is spacing them out, take your protriptyline first and enjoy carob about 30 to 60 minutes later. If you use carob often, just mention it to your pharmacist or doctor.

From our pharmacist’s overview on the Protriptyline page
“Protriptyline (Vivactil) is an older tricyclic antidepressant, and I read its supplement list with genuine care because so much of it piles onto what the drug already does. Four of the 93 ingredients on file rate major, and those deserve your attention before anything else.”

About Carob
What is Carob?
- Diarrhea (especially in children)
- Cocoa/chocolate substitute
- Digestive upset
- Acid reflux/heartburn
- Source of dietary fiber
Carob is a Mediterranean tree whose pods are widely used as a caffeine-free, naturally sweet cocoa alternative and as a thickener (carob gum). It is mostly eaten as a food and is generally well tolerated, and carob pod preparations have some traditional and limited research use for diarrhea, but evidence for most medicinal claims is limited. Check with your pharmacist or doctor before using it as a treatment, especially for infants or children.

Protriptyline Is Linked to Lower Levels of 3 Nutrients
Natural Medicines records Protriptyline as a medicine that can lower these. This is not an interaction with Carob — it is a separate effect of the medicine, and it is a reason some people are advised to supplement rather than to avoid one:
