Lapatinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Clove interacts with.

Theoretically, concomitant use of clove may increase levels of drugs metabolized by CYP3A4.
In vitro research shows that eugenol, the principal constituent of clove, can inhibit CYP3A4 in a dose-dependent manner. This effect has not been reported in humans.

Here's the gist for Tykerb (lapatinib). Your body relies on a liver enzyme called CYP3A4 to break this medication down. Clove's main compound, eugenol, has been shown in lab dish studies to slow that same enzyme. If that happened in real people, lapatinib could build up higher than intended, which might mean more of its known side effects, things like diarrhea, nausea, hand and foot skin reactions, fatigue, or in rare cases effects on heart rhythm or liver function.

I want to be honest about the strength of this: the evidence is only from lab and animal testing, not from humans, so it's theoretical right now. A pinch of clove in cooking is very unlikely to matter. Concentrated clove oil or capsules are a different story. Since lapatinib is a serious cancer drug, please run any clove supplement past your oncologist or pharmacist first.

About Lapatinib

What Is Lapatinib Used For?

Brand name: Tykerb

Lapatinib tablets are used in combination with other medicines to treat certain types of advanced or metastatic breast cancer in patients whose tumors overexpress HER2 (a protein that drives tumor growth). It is not used alone.

Specifically, lapatinib tablets are used with capecitabine for HER2-positive advanced or metastatic breast cancer in patients who have already received prior therapy (including certain chemotherapy drugs and trastuzumab). Lapatinib tablets are also used with letrozole for postmenopausal women with hormone receptor-positive, HER2-positive metastatic breast cancer when hormonal therapy is appropriate. Tykerb is the brand name product with these same indicated uses.
