Abemaciclib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Eleuthero interacts with.

Theoretically, eleuthero might increase levels of drugs metabolized by CYP3A4.
In vitro and animal research suggest that standardized extracts of eleuthero might inhibit CYP3A4. However, research in healthy human volunteers has found that taking eleuthero 485 mg twice daily for 14 days does not inhibit CYP3A4 drug metabolism.

Here's the thinking behind this pairing. Your body clears abemaciclib (Verzenio) using a liver enzyme called CYP3A4. Eleuthero showed some ability to block that enzyme in lab dishes and animal studies, and if it did the same in people, abemaciclib levels could creep up. With this drug that might mean more diarrhea, fatigue, nausea, or low blood counts.

The reassuring part: when researchers actually gave eleuthero to healthy people for two weeks, it did not meaningfully slow CYP3A4. So this concern stays mostly theoretical and is rated minor and unlikely. Still, abemaciclib is a serious cancer medicine, so loop in your oncologist or pharmacist before adding eleuthero rather than starting it on your own.

About Abemaciclib

What Is Abemaciclib Used For?

Brand name: Verzenio

Verzenio (abemaciclib) is used to treat HR-positive, HER2-negative breast cancer in adults. For early breast cancer at high risk of recurrence, Verzenio is taken alongside endocrine therapy (tamoxifen or an aromatase inhibitor) after surgery to help prevent the cancer from coming back.

For advanced or metastatic breast cancer (cancer that has spread), Verzenio can be used in combination with an aromatase inhibitor as a first hormonal treatment, in combination with fulvestrant after the cancer has progressed on endocrine therapy, or alone after prior endocrine therapy and chemotherapy have stopped working.
