First things first: please don't take European mandrake in any form. It's a genuinely toxic plant, and that risk far outweighs anything we discuss about varenicline (Chantix) here.

That said, here's the idea on file. Mandrake's tropane alkaloids slow down how fast your gut moves things along. When your stomach and intestines empty more slowly, a pill like varenicline can sit there longer and get absorbed a bit more fully. In theory, that could nudge your varenicline levels up and make its common side effects, think nausea, vivid or strange dreams, trouble sleeping, and headache, more noticeable. This is theoretical, based on how the plant works rather than real cases, so don't panic. Just loop in your pharmacist or doctor before mixing anything new with varenicline.
