Chlorpheniramine falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Guggul interacts with.
In vitro research shows that guggul constituents known as guggulsterones can induce CYP3A4.
Interaction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports..
Here's the gist for chlorpheniramine: your liver uses an enzyme called CYP3A4 to help break it down. Lab and animal studies suggest the active compounds in guggul (guggulsterones) may rev that enzyme up. If that happens in people, your body could clear chlorpheniramine a bit faster, which might leave you with less allergy relief, so you may still notice sneezing, a runny nose, or itchy, watery eyes.
I'd put this in the 'worth knowing, not worth panicking over' bucket. The evidence is theoretical and comes from test-tube work, not real patients, and chlorpheniramine isn't a high-stakes drug. Still, if you take guggul regularly and your allergy symptoms creep back, mention it to your pharmacist or doctor before changing anything.
