Diazepam falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Horny Goat Weed interacts with.

Theoretically, horny goat weed might increase the effects and side effects of CYP1A2 substrates.
In vitro, horny goat weed leaf extract inhibits CYP1A2. This effect has not been reported in humans.

Here's the gist for Valium (diazepam) and horny goat weed. In lab dish studies, this herb appears to block a liver enzyme called CYP1A2, which is one of the pathways that helps clear certain drugs. Diazepam is an active drug (not a prodrug), so if that enzyme were slowed down, your body could clear it a little slower and the drug might build up. That could mean more of diazepam's usual effects, like extra drowsiness, dizziness, mental fog, or feeling unsteady on your feet.

That said, I don't want to scare you. This is only a theoretical concern seen in test tubes, and it has not been shown in real people. Diazepam is also mainly handled by other enzymes (CYP2C19 and CYP3A4), so CYP1A2 is a smaller player here. Still, since both can make you sleepy, check with your pharmacist or doctor before combining them.

In vitro, horny goat weed extract inhibits CYP3A4 and suppresses CYP3A4 mRNA expression. This effect has not been reported in humans.

Here's the gist: your body relies partly on an enzyme called CYP3A4 to break down diazepam (Valium). In test-tube studies, horny goat weed seems to slow that enzyme down. If that happened in real life, diazepam could clear from your body more slowly and build up, which might mean stronger or longer effects like extra drowsiness, dizziness, foggy thinking, or feeling unsteady on your feet.

I want to be honest about the evidence, though. This has only been seen in lab dishes, not in actual people, so it's a theoretical concern rather than something that's been proven to cause problems. Still, since both can make you sleepy, it's worth a quick chat with your pharmacist or doctor before combining them, especially if you drive or already feel groggy.

What Is Diazepam Used For?

  • Diazepam is used to relieve anxiety and to control agitation caused by alcohol withdrawal. It is also used along with other medications to control muscle spasms and spasticity caused by certain neurological disorders such as cerebral palsy (condition that causes difficulty with movement and balance), paraplegia (inability to move parts of the body), athetosis (abnormal muscle contractions), and stiff-man syndrome (a rare disorder with muscle rigidity and stiffness). Diazepam is also used along with other medications to control seizures. Diazepam is in a class of medications called benzodiazepines. It works by calming abnormal overactivity in the brain.
