Amitriptyline falls under ANTICHOLINERGIC DRUGS — the group Marjoram interacts with.

In vitro research suggests that marjoram extract can inhibit acetylcholinesterase activity. Theoretically, using marjoram in medicinal amounts along with anticholinergic drugs might decrease the effectiveness of marjoram or the anticholinergic agent.
Some anticholinergic drugs include atropine, benztropine (Cogentin), biperiden (Akineton), procyclidine (Kemadrin), and trihexyphenidyl (Artane).

Here is the gist with marjoram and amitriptyline. One of the ways amitriptyline works in the body is by blocking acetylcholine, a nerve signaling chemical. That is the same activity that causes its familiar side effects like dry mouth, blurry vision, constipation, and grogginess. Lab studies hint that marjoram may push acetylcholine levels in the other direction, so in theory the two could partly cancel each other out.

The honest truth is this is theoretical, based only on test-tube work, not real patients. The pinch you cook with almost certainly does not matter. Concentrated marjoram capsules or essential oils are a different story, so just check with your pharmacist or doctor before adding them.
