Naproxen, Sumatriptan falls under ORGANIC ANION TRANSPORTER 3 (OAT3) SUBSTRATES — the group Quercetin interacts with.

Theoretically, concomitant use might increase the effects and adverse effects of OAT3 substrates.

In vitro research shows that quercetin is a strong non-competitive inhibitor of OAT3, with half-maximal inhibitory concentration (IC50) values as low as 0.75 mcM. So far, this interaction has not been reported in humans.

Treximet combines naproxen (an anti-inflammatory pain reliever) and sumatriptan (a migraine medicine). Your kidneys help clear these from your body using a transporter called OAT3. In lab tests, quercetin blocks that transporter. The theory is that if quercetin slows this clearing process, a bit more of the drug could linger in your system, which might nudge up its effects and side effects.

For naproxen that could mean more stomach upset, heartburn, or fluid retention; for sumatriptan, possibly more drowsiness, flushing, or that tight, heavy feeling in the chest or neck. Honestly though, this is only seen in test tubes so far, never reported in real people, and food amounts of quercetin are unlikely to matter. If you take high-dose supplements, just run it by your pharmacist first.

Treximet combines naproxen (an anti-inflammatory) with sumatriptan (a migraine medicine). In lab dish studies, quercetin appears to block that doorway. If that happened in people, a bit more naproxen could linger, which might mean slightly stronger effects or more of its usual side effects like stomach upset, heartburn, or, with heavier use, kidney strain.

Here's the honest part: this is only seen in test-tube research, never reported in actual patients. Food amounts of quercetin almost certainly don't matter. With concentrated supplements, it's still mostly theoretical. If you take high-dose quercetin regularly, just mention it to your pharmacist or doctor.

CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES

Naproxen, Sumatriptan also falls under CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES.

Theoretically, concomitant use might increase the levels and adverse effects of CYP2C9 substrates.

A small clinical study in healthy volunteers shows that taking quercetin 500 mg twice daily for 10 days prior to taking diclofenac, a CYP2C9 substrate, increases diclofenac plasma levels by 75% and prolongs the half-life by 32.5%. Animal research also shows that pretreatment with quercetin increases plasma levels and prolongs the half-life of losartan (Cozaar), a substrate of CYP2C9. Furthermore, laboratory research shows that quercetin inhibits CYP2C9.

Treximet combines two medicines: naproxen (an anti-inflammatory) and sumatriptan (for migraines). The piece that matters here is the naproxen part. Your body relies on a liver enzyme called CYP2C9 to help break naproxen down, and quercetin in concentrated supplement doses can slow that enzyme. When the enzyme works slower, naproxen can hang around longer and build up a bit.

In plain terms, that could nudge up naproxen's usual side effects, things like stomach upset, heartburn, a higher risk of stomach irritation or bleeding, and fluid retention. The evidence here is fairly limited and partly theoretical, drawn from similar drugs, so the everyday amounts of quercetin in foods like apples and onions are unlikely to cause trouble. Before adding a high-dose supplement, just run it by your pharmacist or doctor.
