Moderate evidence
Evidence for Intended Use · database check
By FDA rules, dietary supplements can’t claim to treat, cure, or prevent disease — so labels speak in careful marketing language. We discern each product’s intended use from its name, label claims, and label statements, then grade the clinical evidence for that use. How these ratings are computed
Moderate

Some clinical evidence supports this product's ingredients for its stated purpose, but it isn't conclusive.

Why this rating?
  • The label markets this product for: Joint pain and inflammation support.
  • We looked for evidence on: Osteoarthritis, Rheumatoid arthritis (RA), Knee pain.
  • The strongest evidence on file: Boswellia Serrata is rated "Possibly Effective" for Osteoarthritis (Natural Medicines).
  • Also on file: Boswellia Serrata is rated "Insufficient Reliable Evidence To Rate" for Rheumatoid arthritis (RA), Knee pain.

The evidence for 5-Loxin is strongest for osteoarthritis, where it's rated possibly effective — meaning there's some clinical support, though not yet conclusive. For allergic rhinitis (hay fever), Alzheimer disease, asthma, and cluster headache, the reliable evidence is insufficient to rate whether it works.

The data we hold doesn't establish its effectiveness for other uses.

How safe is it?

Well-documented data
Safety Information · database check
Well characterized

Adverse-effect, pregnancy, and general safety data are on file for most of these ingredients.

Why this rating?
  • We hold adverse-effect (side-effect) data for 1 of the 1 matched ingredient.
  • Pregnancy & breastfeeding safety ratings cover 1 of 1.
  • General safety write-ups exist for 1 of 1.
  • Remember: this measures how much safety information exists. Thin data is not the same as being safe.

Boswellia serrata taken by mouth is generally well tolerated in the short term, though quality and dose vary widely between products — so consistency matters. The most common side effects are mild: abdominal pain, diarrhea, headache, heartburn, itching, and nausea.

In one clinical study, nearly 11% of people taking a specific Boswellia extract reported headache; dizziness and vertigo were also reported at lower rates. Rarely, very large amounts of the gum resin can cause a bezoar (a mass of plant material blocking the gut).

During pregnancy, there isn't enough safety data, so it's best avoided. While breastfeeding, safety is unknown, so it's best to avoid it as well.

Before you take 5-Loxin, check any medications that are metabolized by liver enzymes CYP2D6, CYP2C19, CYP1A2, CYP2C9, or CYP3A4 — this includes many common heart, psychiatric, and pain medications. Also check if you're on immunosuppressants or any immune-modulating drugs.

The bottom line

Scorecard at a glanceFully disclosed formula with some supporting evidence for its stated purpose. Moderate medication interactions have been identified, and safety information is well characterized.

5-Loxin may be worth considering if you have osteoarthritis and want to try a botanical approach, but you'll need to check it against any medications you're on — it affects how your liver processes a large number of drugs. If you're on heart, blood pressure, or psychiatric medications, or anything that suppresses your immune system, talk with your pharmacist or doctor before starting.

Cytochrome P450 1A2 (Cyp1A2) Substrates

Theoretically, Boswellia serrata might increase the levels of CYP1A2 substrates.
In vitro research shows that Boswellia serrata gum resin inhibits CYP1A2 enzymes.

Cytochrome P450 2C19 (Cyp2C19) Substrates

Theoretically, Boswellia serrata might increase the levels of CYP2C19 substrates.
In vitro research shows that Boswellia serrata gum resin inhibits CYP2C19 enzymes.

Likelihood Possible Evidence D
Cytochrome P450 2C9 (Cyp2C9) Substrates

In vitro research shows that Boswellia serrata gum resin inhibits CYP2D6 enzymes.

Likelihood Possible Evidence D
Cytochrome P450 3A4 (Cyp3A4) Substrates

Theoretically, Boswellia serrata might increase or decrease the levels and clinical effects of CYP3A4 substrates.
In vitro research shows that Boswellia serrata gum resin inhibits CYP3A4 enzymes. Other in vitro research shows that Boswellia serrata extract inhibits CYP3A4 enzymes at most concentrations, although it may modestly induce enzyme activity at low concentrations.

Likelihood Possible Evidence D
Immunosuppressants

Theoretically, Boswellia serrata might alter the effects of immunosuppressive drugs.
Some in vitro research suggests that Boswellia serrata extracts might inhibit mediators of autoimmune disorders such as leukotrienes and reduce production of antibodies and cell-mediated immunity. However, other in vitro research suggests that, when coupled with calcium ions, boswellic acids containing the keto group have immunostimulant properties within specific cell signaling pathways.

How can one product interact with so many drugs?
5-Loxin 200 mg contains a single active ingredient, and that one ingredient can interact with many different medications on its own. We check it against each medication and show the mechanism responsible.
Where does this information come from?
The product label data comes from the NIH Dietary Supplement Label Database (DSLD); the interaction data is built on the Natural Medicines database and reviewed by HelloPharmacist pharmacists.
