Enzaid is a 12-ingredient digestive enzyme blend. The active components include papain (a protein-digesting enzyme from papaya), diastase (which breaks down starches), pepsin (another protein digestor), lipase (which breaks down fats), and pancreatin (a pancreatic enzyme extract), along with L-glutamic acid hydrochloride, beet, citrus pectin cellulose, ox bile extract, betaine, mycozyme, and rennin.
The product also contains inactive ingredients—stearic acid, microcrystalline cellulose, croscarmellose sodium, silicon dioxide, magnesium stearate, guar gum, hydroxypropyl cellulose, hypromellose, and glycerin—which are fillers, binders, and capsule materials.

The evidence for Enzaid's active ingredients is limited. Papain has insufficient reliable evidence for cancer, dental caries, radiation skin damage, exercise-related muscle soreness, shingles, and intestinal parasite infection.
Beet shows possibly effective evidence for athletic performance and exercise-induced muscle soreness, but possibly ineffective for COPD, and insufficient evidence for cholesterol levels and mental function. Pepsin, lipase, and the other ingredients in this blend lack established effectiveness ratings in our data.
How safe is it?
Well-documented data
Papain is generally tolerated as a food but may irritate the digestive tract and cause allergic reactions in sensitive people—itchy watery eyes, runny nose, sneezing, abdominal cramps, sweating, or diarrhea. High doses may be unsafe and rarely can cause esophageal perforation or severe stomach inflammation.
Papain is possibly unsafe in pregnancy and lacks enough data to recommend during breastfeeding. Beet is widely eaten as food and generally well tolerated, though concentrated supplements lack safety data; most common effects are red or black stools (harmless), and rarely, low calcium levels or kidney damage with large amounts.
Pepsin is generally well tolerated but may cause digestive upset or allergic reactions; safety in pregnancy has not been established, and medical advice is needed before breastfeeding. Lipase is generally well tolerated but should be used only under medical guidance if you have serious digestive conditions; safety in pregnancy and breastfeeding has not been well studied.

Also confirm your use if you're on blood pressure medications (beet may lower pressure), or any drugs processed through your liver's CYP3A4 enzymes—a large group that includes many statins, antiarrhythmics, and immunosuppressants.

The most serious is papain's interaction with warfarin (Coumadin), a blood thinner—papain may increase warfarin's effects and side effects, raising bleeding risk. One case report documented a patient stable on warfarin whose blood-clotting measure (INR) jumped to 7.4 after taking a papaya extract supplement containing papain.

Beet
Cytochrome P450 3A4 (Cyp3A4) Substrates
Theoretically, beet might increase the levels of CYP3A4 substrates.
In vitro research suggests that betanin, the major pigment in beet, competitively inhibits CYP3A4 in a dose-dependent manner similarly to strong CYP3A4 inhibitor ketoconazole.
Antihypertensive Drugs
Beet and beetroot contain nitrates, which can cause vasodilation, potentially leading to lower blood pressure. However, a study published in the European Journal of Clinical Nutrition using concentrated beetroot juice found no significant impact on blood pressure or heart rate in different age groups. Other small clinical studies suggest that while beet consumption might transiently lower blood pressure due to vessel dilation, there's no consistent evidence of a lasting effect. Overall, the theoretical risk of reduced blood pressure due to beet's nitrate content exists, but studies generally indicate a low and temporary impact rather than a sustained decrease.
Cytochrome P450 1A2 (Cyp1A2) Substrates

Warfarin (Coumadin)
Theoretically, papain might increase the effects and side effects of warfarin.
In one case report, a patient previously stable on warfarin was found to have an international normalization ratio (INR) of 7.4, which was attributed to ingestion of a supplement containing papain from papaya extract.
