L-glutamine is generally well tolerated in healthy adults when taken by mouth. The most common side effects are gastrointestinal — belching, bloating, constipation, diarrhea, flatulence, nausea, vomiting, and stomach pain — especially at higher doses (10–30 grams daily or higher).

Headache, dizziness, and musculoskeletal pain have also been reported. A very small number of people with bipolar disorder taking high doses (up to 4 grams daily) have reported mania or hypomania.
