Acacia Rigidula + Imipramine
Imipramine brand names: Tofranil, Tofranil PM.
Imipramine falls into 2 drug categories that Acacia Rigidula can interact with; the most serious is rated moderate. Theoretically, Acacia rigidula products that are adulterated with beta-methylphenylethylamine (BMPEA) might increase the levels of CYP2D6 substrates.

These two interact in 2 distinct ways. Each one is explained separately below.
Why Are There 2 Interactions?
Acacia Rigidula interacts with whole categories of medication. Imipramine falls into 2 of those categories, so it can interact with Acacia Rigidula in 2 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).
CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES
1 of 2Why this applies to Imipramine
Imipramine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Acacia Rigidula interacts with.
In vitro research shows that BMPEA, an adulterant found in many Acacia rigidula products, strongly inhibits CYP2D6 enzyme.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with imipramine: your liver leans heavily on an enzyme called CYP2D6 to break this medicine down and clear it out. Some Acacia rigidula products have been found spiked with a hidden stimulant called BMPEA, and lab studies suggest BMPEA can block that very enzyme. If it's blocked, imipramine could pile up in your system instead of being cleared at the usual pace.
More imipramine than expected can mean stronger side effects like a fast or pounding heartbeat, dizziness, dry mouth, constipation, blurred vision, and drowsiness. This is a theoretical concern based only on lab work, not proven in people, so I'm not trying to scare you. Still, since imipramine affects the heart and the supplement adds stimulant effects, please check with your pharmacist or doctor before combining them.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
2 of 2Imipramine also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.
In vitro research shows that BMPEA, an adulterant found in many Acacia rigidula products, inhibits CYP3A4 enzyme by 37%.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with imipramine (Tofranil). Some Acacia rigidula products have been found spiked with a stimulant called BMPEA, and in lab dish studies that compound slowed down a liver enzyme called CYP3A4 by about a third. Imipramine is partly cleared by that same enzyme, so in theory blocking it could let imipramine build up a bit higher in your body.
If that happened, you might notice more of the usual tricyclic side effects: dry mouth, constipation, dizziness, a faster heartbeat, or drowsiness. I'll be honest though, this is theoretical and based only on test-tube work, not real people. On top of that, the stimulant amines in these products can stack with imipramine on the heart and blood pressure. I'd skip this supplement and run it by your pharmacist or doctor first.

From our pharmacist’s overview on the Imipramine page
“Imipramine (Tofranil) is an older tricyclic antidepressant, and the supplements on this page deserve real scrutiny before you add one. Most of the list acts on the liver enzymes that process the drug, so an ingredient can quietly raise or lower imipramine levels: grapefruit pushes them up, while St.”

About Acacia Rigidula
What is Acacia Rigidula?
- Weight loss
- Energy and stimulation
- Boosting metabolism
- Appetite control
- Pre-workout/athletic performance
Acacia rigidula is a shrub from Texas and Mexico that is marketed in weight-loss and energy supplements. There is very little reliable human evidence that it works, and some products have been found to contain an unapproved stimulant drug (BMPEA), which raises real safety concerns. Most people should avoid it and talk to a pharmacist or doctor before trying any product containing it.
About Imipramine
What Is Imipramine Used For?
Brand names: Tofranil, Tofranil PM
Imipramine is used to relieve symptoms of depression. Both Imipramine Hydrochloride tablets and Imipramine Pamoate capsules are approved for this use. Endogenous depression — the kind that seems to arise from within rather than a clear outside cause — tends to respond especially well. It may take one to three weeks before you notice the full benefit.
Imipramine Hydrochloride tablets are also approved as a short-term, add-on treatment to help reduce nighttime bedwetting (nocturnal enuresis) in children aged 6 and older, after a doctor has ruled out physical causes.
