Vortioxetine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Berberine interacts with.

Theoretically, berberine might increase serum levels of drugs metabolized by CYP2D6.
In vitro research and preliminary clinical evidence show that berberine can inhibit CYP2D6. Theoretically, use of berberine with drugs metabolized by CYP2D6 might increase drug levels and increase the risk of adverse effects.

Here's the simple version: your body relies heavily on a liver enzyme called CYP2D6 to clear vortioxetine (Trintellix) out of your system. Berberine appears to slow that enzyme down a bit. When the cleanup crew works slower, more of the drug can hang around, which may nudge its effects and side effects up. For vortioxetine, that could mean more nausea, headache, dizziness, or trouble sleeping.

I want to keep this in perspective, though. This concern is mostly theoretical, based on lab work and only early, lower quality human studies. It's a possible interaction, not a sure thing. Vortioxetine is not a prodrug, so berberine wouldn't weaken it. Before adding berberine, just run it by your pharmacist or doctor so they can keep an eye on how you feel.
