Here is the interaction we found between these two, with the evidence behind it.
CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES
Why this applies to Ramelteon
Ramelteon falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Black Hoof Mushroom interacts with.
In vitro, polysaccharides from black hoof mushroom grown in culture inhibit CYP1A2. This effect has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for Ramelteon: your body clears it mainly through a liver enzyme called CYP1A2. In lab dish studies, compounds from Black Hoof Mushroom seemed to slow that same enzyme down. If that were to happen in real life, more Ramelteon could hang around in your system, which might leave you feeling extra drowsy, groggy, or dizzy the next morning, or make it harder to wake up fully.
I'll be straight with you, though: this concern comes only from test-tube research, not from people, so it's theoretical at this point. Ramelteon isn't a prodrug, so the direction would be stronger effects, not weaker. Still, before you pair these two, give your pharmacist or doctor a quick heads-up so they can keep an eye on you.

“Ramelteon (Rozerem) is a prescription sleep aid, and its 228 interacting supplements call for real care, starting with the nine rated major. The frustrating part is that the things sleepless people typically reach for, kava, CBD, a glass of wine before bed, sit at the heart of the problem.”
