Why this applies to Entrectinib

Entrectinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Clove interacts with.

Theoretically, concomitant use of clove may increase levels of drugs metabolized by CYP3A4.
In vitro research shows that eugenol, the principal constituent of clove, can inhibit CYP3A4 in a dose-dependent manner. This effect has not been reported in humans.

Here's the short version: your body clears entrectinib (Rozlytrek) mainly through a liver enzyme called CYP3A4. In lab dishes, eugenol (the main compound in clove) has been shown to slow that same enzyme down. If that happened in people, entrectinib could clear more slowly and build up, which might mean stronger side effects like dizziness, fatigue, swelling, constipation, or changes in your heart rhythm.

The important caveat: this is based only on test-tube research, not real patients, so it's theoretical. And the clove in your cooking is very unlikely to matter. Concentrated clove oil or clove supplements are the bigger question mark. Since entrectinib is a serious cancer medicine, please run any clove products by your oncology pharmacist or doctor first.

What Is Entrectinib Used For?

Brand name: Rozlytrek

Rozlytrek is used to treat adult patients with ROS1-positive metastatic non-small cell lung cancer (NSCLC) — a type of lung cancer where the ROS1 gene is rearranged — as confirmed by an FDA-approved test.

Rozlytrek is also approved for adult and pediatric patients older than 1 month with solid tumors that have an NTRK gene fusion (a specific genetic alteration), detected by an FDA-approved test, that have spread or cannot be removed without causing serious harm, and have worsened after prior treatment or have no good treatment alternatives. This NTRK indication was granted accelerated approval based on tumor response rates; continued approval may depend on results from confirmatory trials.
