Acetaminophen, Orphenadrine falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Evodia interacts with.
Evodia extract and the evodia constituent rutaecarpine induce hepatic CYP1A2 enzyme activity. Evodia decreases levels of theophylline and caffeine, CYP1A2 substrates, by about 70% in animal models.
Interaction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports..
This combination pill pairs acetaminophen (a pain and fever reducer) with orphenadrine (a muscle relaxant). Evodia is thought to speed up a liver enzyme called CYP1A2, which can help clear certain drugs out of your body faster. In theory, that could mean a little less benefit from this medication, so your aches, muscle tightness, or fever might not be controlled as well.
Here's the important part: this is based only on test tube and animal research, not on people, so we honestly don't know how much it matters in real life. I wouldn't lose sleep over it, but it's worth a quick chat with your pharmacist or doctor before adding concentrated Evodia supplements, especially so they can keep an eye on whether your pain relief still feels effective.

CYTOCHROME P450 2E1 (CYP2E1) SUBSTRATES

Animal research suggests that rutaecarpine, a constituent of evodia, induces CYP2E1 activity. In rats, rutaecarpine increases markers of CYP2E1 activity, and administration of rutaecarpine with chlorzoxazone, a known CYP2E1 substrate, reduces the area under the curve (AUC) of chlorzoxazone by 84% and increases its clearance by 646%.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for this combo pill (acetaminophen plus orphenadrine). The acetaminophen part is partly handled by a liver enzyme called CYP2E1. Evodia, thanks to a compound called rutaecarpine, may speed that enzyme up. In theory, faster processing could mean your body clears acetaminophen a bit quicker, so it might not relieve your pain or fever quite as well.
There's an important catch with acetaminophen, though. CYP2E1 is the same pathway that turns it into a toxic byproduct, and revving up that route can mean more strain on the liver, especially at higher doses or with alcohol. I'll be honest: this is based only on animal lab work, so it's theoretical and unproven in people. Still, please run it by your pharmacist or doctor before pairing them.
