Ponatinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Horny Goat Weed interacts with.

Theoretically, horny goat weed might increase the effects and side effects of CYP3A4 substrates.
In vitro, horny goat weed extract inhibits CYP3A4 and suppresses CYP3A4 mRNA expression. This effect has not been reported in humans.

Here's the gist: your body relies on a liver enzyme called CYP3A4 to clear ponatinib (Iclusig). In test-tube studies, horny goat weed seems to slow that same enzyme down. If that happened in people, ponatinib could build up in your blood and hit harder than intended. With ponatinib, that's a real concern because its known risks include blood clots, high blood pressure, liver problems, and serious heart issues, so more drug in your system could mean more of those side effects.

The honest part: this is based only on lab work, not on real patients, so we can't say how strong it would be in you. Still, ponatinib is a powerful cancer medicine, so please check with your oncologist or pharmacist before adding this supplement.

About Ponatinib

What Is Ponatinib Used For?

Brand name: Iclusig

Iclusig (ponatinib) is used to treat adult patients with Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL) — either newly diagnosed (in combination with chemotherapy) or in patients for whom no other kinase inhibitors are appropriate, or who have a specific mutation called T315I. It is also used for adults with chronic myeloid leukemia (CML), including chronic phase, accelerated phase, and blast phase CML, in situations where prior treatments have not worked or are not tolerated, or where the T315I mutation is present.

Iclusig is not recommended for patients newly diagnosed with chronic phase CML who have not yet tried other treatments.
