L-tryptophan and Estazolam

Here is the interaction we found between these two, with the evidence behind it.

Major

CNS DEPRESSANTS

Why this applies to Estazolam

Estazolam falls under CNS DEPRESSANTS — the group L-tryptophan interacts with.

Theoretically, concomitant use of L-tryptophan with CNS depressants might cause additive sedative effects.
Clinical research shows that L-tryptophan can cause fatigue and drowsiness.
Severity Major Likelihood Probable Evidence B

Interaction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports..

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the thing to keep in mind: Estazolam (Prosom) is a benzodiazepine sleeping pill that already slows down your brain to help you fall asleep. L-tryptophan isn't broken down by the same liver pathways, so this isn't about one changing the other's blood levels. Instead, the concern is simple addition. Tryptophan raises serotonin and can make you feel drowsy and tired on its own, and Estazolam does the same. Stack them together and that sleepy, sedated effect can pile up.

In real life, that could mean feeling extra groggy, dizzy, unsteady on your feet, or having a harder time waking up clearly the next morning. The evidence here is fairly suggestive rather than rock-solid, but since both are aimed at sleep, please check with your pharmacist or doctor before combining them.

About Estazolam

What Is Estazolam Used For?

Brand name: Prosom

Estazolam tablets are used for the short-term management of insomnia — specifically trouble falling asleep, waking up frequently during the night, and waking too early in the morning.

Because insomnia is often temporary, long-term use of estazolam is generally not recommended. If sleep problems last beyond 7–10 days of treatment, a doctor should evaluate whether an underlying medical or psychiatric condition is causing them.
