Interaction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports..

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the simple version: both L-tryptophan and Ingrezza (valbenazine) can make you feel sleepy on their own. Valbenazine commonly causes drowsiness, and tryptophan supplements have been shown to bring on fatigue and drowsiness too. Put them together and those tired, foggy feelings may stack up, so you could feel more sleepy, slowed down, or less alert than usual, especially when driving or operating machinery.

This is mostly an additive effect on the brain, not a change in how your body breaks the drug down. It's worth a quick chat with your pharmacist or doctor before combining them, particularly when you're starting either one. Don't stop or change your valbenazine on your own.
