Acetaminophen, Chlorzoxazone, Codeine falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Licorice interacts with.

Theoretically, licorice might decrease the levels and clinical effects of CYP1A2 substrates.

This combination pill mixes a pain reliever (acetaminophen), a muscle relaxer (chlorzoxazone), and an opioid (codeine). The interaction on file is about an enzyme called CYP1A2, and the concern is mostly theoretical. Lab and test-tube studies suggest licorice might speed that enzyme up a bit, which in theory could lower the levels of medicines it processes. In real life, the human evidence here is very weak, so I wouldn't lose sleep over it.

One thing worth knowing: codeine is a prodrug, meaning your body has to activate it (through a different enzyme, CYP2D6) to relieve pain. So this particular pathway isn't its main route. A bigger reason to be careful with licorice is that real licorice can raise blood pressure and drop potassium. Check with your pharmacist before pairing them.

Licorice root is a traditional remedy used for sore throats, coughs, and digestive complaints, but solid human evidence is limited for most uses. Regular licorice contains glycyrrhizin, which can raise blood pressure and lower potassium when taken in large amounts or for a long time, so deglycyrrhizinated (DGL) products are often preferred. Talk to your pharmacist or doctor before using it, especially if you have high blood pressure, heart, or kidney problems.
