Why this applies to Acetaminophen
Acetaminophen falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Licorice interacts with.
In vitro research shows that licorice induces CYP1A2 enzymes.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with Tylenol (acetaminophen) and licorice. Some lab studies suggest licorice may speed up an enzyme called CYP1A2, which is one of the pathways your body uses to process acetaminophen. In theory, that could clear the drug a bit faster and make it slightly less effective for your pain or fever. So if anything, you might notice your Tylenol not lasting quite as long.
That said, this is a minor, theoretical concern based only on test-tube and animal research, not real patients. For most people taking occasional Tylenol, it isn't something to worry about. If you use licorice regularly and feel your pain relief is falling short, just check in with your pharmacist or doctor.

Licorice root is a traditional remedy used for sore throats, coughs, and digestive complaints, but solid human evidence is limited for most uses. Regular licorice contains glycyrrhizin, which can raise blood pressure and lower potassium when taken in large amounts or for a long time, so deglycyrrhizinated (DGL) products are often preferred. Talk to your pharmacist or doctor before using it, especially if you have high blood pressure, heart, or kidney problems.

Across the oral products (tablets and suspensions), acetaminophen temporarily relieves minor aches and pains and temporarily reduces fever.
The aches and pains named include the common cold, headache, backache, minor arthritis pain, toothache, muscle aches, and menstrual and premenstrual cramps. Robitussin Direct Sore Throat Pain tablets also list sore throat pain.
