Why this applies to Lofepramine

Lofepramine falls under CYTOCHROME P450 2C19 (CYP2C19) SUBSTRATES — the group Licorice interacts with.

Theoretically, licorice might increase levels of drugs metabolized by CYP2C19.
In vitro, licorice extracts from the species G. glabra and G. uralensis inhibit CYP2C19 isoenzymes in vitro. Theoretically, these species of licorice might increase levels of drugs metabolized by CYP2C19; however, this interaction has not yet been reported in humans.

Here's the gist for Lofepramine, a tricyclic antidepressant. One of the liver enzymes that helps clear it is called CYP2C19. In lab dish studies, licorice (from certain Glycyrrhiza species) appears to slow that enzyme down. If that happened in the body, your Lofepramine could build up a bit more than usual, which might mean stronger side effects like drowsiness, dry mouth, constipation, blurry vision, or feeling lightheaded when you stand.

The important thing: this is only theoretical, based on test-tube work, and it has never actually been reported in people. So I wouldn't lose sleep over it. Still, if you're using concentrated licorice supplements regularly, it's worth a quick chat with your pharmacist or doctor before changing anything.

Theoretically, licorice might decrease the levels and clinical effects of CYP1A2 substrates.
In vitro research shows that licorice induces CYP1A2 enzymes.

Lofepramine is a tricyclic antidepressant, and your liver partly clears it using an enzyme called CYP1A2. The concern here is that licorice (specifically the glycyrrhizin in regular, non-DGL products) might speed up that enzyme. If it does, your body could break down lofepramine a little faster, which in theory means slightly lower levels and a weaker effect on your mood or anxiety.

Honestly, this is a very weak, theory-only worry. It comes from lab studies, not real people, and we've graded it as minor. Most folks won't notice anything. Still, if you take licorice regularly and feel your depression creeping back, mention it to your pharmacist or doctor before changing anything.

Licorice root is a traditional remedy used for sore throats, coughs, and digestive complaints, but solid human evidence is limited for most uses. Regular licorice contains glycyrrhizin, which can raise blood pressure and lower potassium when taken in large amounts or for a long time, so deglycyrrhizinated (DGL) products are often preferred. Talk to your pharmacist or doctor before using it, especially if you have high blood pressure, heart, or kidney problems.
