Modafinil falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Polypodium Leucotomos interacts with.

Theoretically, Polypodium leucotomos might increase levels of drugs metabolized by CYP3A4.
In vitro and animal research suggests that Polypodium leucotomos can decrease the metabolism of midazolam by a mechanism possibly related to the inhibition of CYP3A4. However, a small study in humans suggests that taking Polypodium leucotomos with midazolam does not affect the metabolism of midazolam.

Here's the gist: modafinil is partly cleared by a liver enzyme called CYP3A4. The thinking is that Polypodium leucotomos might slow that enzyme down a touch, which in theory could let a bit more modafinil hang around in your body. If that happened, you might notice modafinil's usual effects a little more strongly, things like jitteriness, headache, a racing heart, trouble sleeping, or feeling on edge.

That said, I want to keep this in perspective. This is rated a minor and unlikely interaction, and it's mostly based on lab and animal work. A small human study with a similar drug actually showed no real change. So for most people this isn't a big worry. Still, it's worth a quick mention to your pharmacist or doctor before you pair them, especially if you notice new side effects.

What Is Modafinil Used For?

Brand names: Provigil, Sparlon, Alertec

Modafinil (also sold as Provigil) is used to improve wakefulness in adults who experience excessive sleepiness due to narcolepsy, obstructive sleep apnea (OSA), or shift work disorder (SWD).

In OSA, modafinil treats the sleepiness — not the underlying airway obstruction itself. It is used alongside standard treatments such as CPAP (a breathing device worn during sleep), not as a replacement for them. If CPAP is appropriate for you, your doctor will want you to use it consistently even while taking modafinil.
