Concomitant use of pregnenolone may reduce the effects of benzodiazepines.
Very preliminary clinical research shows that chronic use of pregnenolone reduces sedative effects of diazepam when compared with chronic use of placebo. This effect may be related to the activity of pregnenolone at GABAA receptors.

Here's the gist for Clobazam (Onfi, Sympazan): both this seizure medicine and pregnenolone work on the same brain switch, the GABA-A receptor. Some early human research suggests that taking pregnenolone regularly may blunt the calming, sedative effect of benzodiazepines like clobazam. In plain terms, the supplement could make your clobazam feel like it's working a little less than usual.

For a medicine you rely on to control seizures, that matters. If clobazam's effect is dampened, there's a theoretical worry about less seizure protection, or simply a change in how drowsy or steady you feel. The evidence here is limited and comes from a small, lower-quality study using a different benzodiazepine, so this isn't a sure thing. Still, please don't add pregnenolone or adjust your clobazam without first checking with your neurologist or pharmacist.

Clobazam is used as an add-on (adjunctive) treatment for seizures linked to Lennox-Gastaut Syndrome (LGS), a severe form of childhood-onset epilepsy. It is approved for patients 2 years of age and older.

This indication applies to all available forms: Sympazan (dissolving oral film), Onfi (tablets and oral suspension), and generic clobazam tablets and oral suspension.
