CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES

Amitriptyline, Chlordiazepoxide falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Quercetin interacts with.
In vitro research show that quercetin inhibits CYP2D6. This effect has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Limbitrol DS combines amitriptyline (an antidepressant) with chlordiazepoxide (a benzodiazepine for anxiety). The amitriptyline part is broken down partly by a liver enzyme called CYP2D6. In test-tube studies, quercetin can slow that enzyme down. If that happened in your body, amitriptyline could build up a little, which might mean more of its usual effects like drowsiness, dry mouth, constipation, blurry vision, dizziness on standing, or a faster heartbeat.
Here is the reassuring part: this has only been seen in lab studies, not in real people, so it is theoretical. Food amounts of quercetin are very unlikely to matter. Still, since this combo is already sedating, if you are considering high-dose quercetin supplements, please run it by your pharmacist or doctor first.

CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES

A small clinical study in healthy volunteers shows that taking quercetin 500 mg twice daily for 10 days prior to taking diclofenac, a CYP2C9 substrate, increases diclofenac plasma levels by 75% and prolongs the half-life by 32.5%. Animal research also shows that pretreatment with quercetin increases plasma levels and prolongs the half-life of losartan (Cozaar), a substrate of CYP2C9. Furthermore, laboratory research shows that quercetin inhibits CYP2C9.

Limbitrol DS combines amitriptyline (an older antidepressant) with chlordiazepoxide (a benzodiazepine for anxiety). The concern here is that quercetin can mildly block a liver enzyme called CYP2C9, which helps process certain medicines. In theory, that could let levels of this combo pill climb a bit higher than usual.
If that happened, you might feel more of the usual effects, like drowsiness, dizziness, dry mouth, constipation, or feeling foggy and unsteady. That said, this is mostly theoretical, and CYP2C9 isn't even the main pathway for these two drugs (they lean more on other enzymes). Food amounts of quercetin are very unlikely to matter. If you're considering a high-dose supplement, just run it by your pharmacist or doctor first so they can keep an eye on you.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES

A small clinical study in healthy volunteers shows that pretreatment with quercetin increases plasma levels and prolongs the half-life of a single dose of cyclosporine (Neoral, Sandimmune), a substrate of CYP3A4. Animal research also shows that pretreatment with quercetin increases plasma levels and prolongs the half-life of losartan (Cozaar) and quetiapine (Seroquel), substrates of CYP3A4. Other laboratory research also shows that quercetin inhibits CYP3A4. However, one clinical study shows that quercetin can increase the metabolism of midazolam, a substrate of CYP3A4, and decrease serum concentrations of midazolam by about 24% in some healthy individuals, suggesting possible induction of CYP3A4.

Here's the deal with Limbitrol DS, which combines amitriptyline (an older antidepressant) and chlordiazepoxide (a benzodiazepine). Both rely partly on a liver enzyme called CYP3A4 to be broken down. In lab and animal studies, quercetin has been shown to block this enzyme, which could in theory let these medicines build up a bit higher than usual. If that happened, you might notice more drowsiness, dizziness, dry mouth, constipation, or feeling extra groggy.
That said, the human evidence here is thin and even a little contradictory, so we don't really know how much this matters in real life. Food amounts of quercetin are very unlikely to cause trouble, but concentrated supplements are worth running by your pharmacist or doctor first.
