Rosiglitazone Maleate falls under CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES — the group Quercetin interacts with.
A small clinical study in healthy volunteers shows that taking quercetin 500 mg twice daily for 10 days prior to taking diclofenac, a CYP2C9 substrate, increases diclofenac plasma levels by 75% and prolongs the half-life by 32.5%. Animal research also shows that pretreatment with quercetin increases plasma levels and prolongs the half-life of losartan (Cozaar), a substrate of CYP2C9. Furthermore, laboratory research shows that quercetin inhibits CYP2C9.

Here's the gist for your Avandia (rosiglitazone). Your liver uses an enzyme called CYP2C9 to help clear this medication out of your body. Quercetin, when taken as a concentrated supplement, can slow that same enzyme down. If it does, rosiglitazone may hang around longer and build up a bit more than usual, which could nudge its blood-sugar-lowering effect and side effects upward. With rosiglitazone, that might look like low blood sugar, fluid retention or swelling in the legs, or some weight gain.
I want to be honest about the evidence here: this is mostly theoretical, based on lab work and studies with other drugs, not rosiglitazone itself. Everyday food amounts of quercetin are unlikely to matter. If you're considering high-dose supplements, just run it by your pharmacist or doctor first so they can keep an eye on your numbers.

Clinical research suggests that a combination of quercetin, myricetin, and chlorogenic acid reduce levels of fasting glucose in patients with type 2 diabetes, including those already taking antidiabetes agents. The effect of quercetin alone is unknown.

Here's the gist for rosiglitazone (Avandia): this medicine already lowers your blood sugar by making your body more sensitive to insulin. Quercetin may have a mild blood-sugar-lowering effect of its own, so taking a concentrated quercetin supplement on top of your rosiglitazone could, in theory, nudge your sugar down a little further than expected. If that happens, you might notice signs of low blood sugar (hypoglycemia) like shakiness, sweating, dizziness, or feeling unusually hungry or foggy.
I want to be honest that this is mostly theoretical. The clearest research used quercetin combined with other compounds, and quercetin's solo effect isn't well proven. The amounts in everyday foods are very unlikely to matter. If you're considering a high-dose supplement, check with your pharmacist or doctor first, and keep an eye on your home glucose readings.

CYTOCHROME P450 2C8 (CYP2C8) SUBSTRATES

In vitro research shows that quercetin inhibits CYP2C8. Inhibition of paclitaxel (Taxol) metabolism via CYP2C8 has been reported in vitro. However, a small study in humans found no effect of quercetin on rosiglitazone (Avandia), which is also a CYP2C8 substrate.

Here's the gist: your body clears rosiglitazone (Avandia) using a liver enzyme called CYP2C8. In test-tube studies, quercetin can slow that same enzyme down. On paper, that could let rosiglitazone build up a bit, which in theory might mean stronger blood-sugar lowering or side effects like swelling (fluid retention), weight gain, or feeling tired.
But here's the reassuring part: this concern comes from lab research, not real people. In fact, a small human study found that quercetin did not actually change rosiglitazone levels. So this is mostly theoretical, and rosiglitazone is not a prodrug, so it doesn't flip on us. If you take both, it's still smart to keep an eye on your blood sugar and mention it to your pharmacist or doctor, but you don't need to panic.

Rosiglitazone is used along with a diet and exercise program and sometimes with one or more other medications to treat type 2 diabetes (condition in which the body does not use insulin normally and therefore cannot control the amount of sugar in the blood). Rosiglitazone is in a class of medications called thiazolidinediones. It works by increasing the body's sensitivity to insulin, a natural substance that helps control blood sugar levels. Rosiglitazone is not used to treat type 1 diabetes (condition in which the body does not produce insulin and therefore cannot control the amount of sugar in the blood) or diabetic ketoacidosis (a serious condition that may occur if high blood sugar is not treated).

Over time, people who have diabetes and high blood sugar can develop serious or life-threatening complications, including heart disease, stroke, kidney problems, nerve damage, and eye problems. Taking medication(s), making lifestyle changes (e.g., diet, exercise, quitting smoking), and regularly checking your blood sugar may help to manage your diabetes and improve your health. This therapy may also decrease your chances of having a heart attack, stroke, or other diabetes-related complications such as kidney failure, nerve damage (numb, cold legs or feet; decreased sexual ability in men and women), eye problems, including changes or loss of vision, or gum disease.
