L-Glutamine is generally well tolerated by healthy adults. The most common side effects from oral glutamine are mild and digestive: belching, bloating, constipation, flatulence, diarrhea, nausea, vomiting, and gastrointestinal pain.

Headache and musculoskeletal pain have also been reported. A few rare cases of dizziness, and in two patients with bipolar disorder, mania or hypomania, have been reported — though in one dizziness case, the symptom went away when the dose was lowered.
