The evidence for L-tryptophan's usefulness is mixed and limited. For depression, the data suggests it's possibly ineffective.
For anxiety, athletic performance, bruxism (teeth grinding), and premenstrual dysphoric disorder, we don't have enough reliable evidence to know whether it works.
How safe is it?
Well-documented data
L-Tryptophan is generally well tolerated by mouth, though common side effects include belching, diarrhea, drowsiness, dry mouth, flatulence, headache, heartburn, lightheadedness, nausea, stomach pain, blurred vision, and vomiting. Importantly, L-tryptophan has a serious historical safety concern: in 1989, more than 1,500 cases of eosinophilia-myalgia syndrome (EMS)—a rare neurological disorder with skin changes, hair loss, and rash—were linked to contaminated L-tryptophan from a single Japanese manufacturer.
That led to an FDA recall and import limits in 1990. Almost all cases traced to that contamination.
The safety data advises against use during pregnancy and while breastfeeding. If you're pregnant, planning pregnancy, or nursing, talk with your doctor or pharmacist before considering this supplement.
